[225Ac]PrDOTA-Bn is a radiolabeled hapten designed for use in pretargeted radioimmunotherapy (PRIT), specifically within the 'proteus-DOTA' (Pr-DOTA) alpha-particle therapy platform. It consists of the alpha-emitting radionuclide Actinium-225 (225Ac) chelated by a specialized dual-DOTA construct (PrDOTA-Bn) intended to improve the stability and delivery of the radioisotope compared to standard DOTA-Bn haptens. In the PRIT workflow, a bispecific antibody (BsAb) is first administered to bind tumor-associated antigens (such as HER2, GPA33, or GD2). After the BsAb has cleared from the blood, [225Ac]PrDOTA-Bn is injected; it rapidly binds to the pre-localized BsAb via an anti-DOTA single-chain variable fragment (scFv), delivering high-LET alpha radiation directly to the tumor site while minimizing systemic exposure. This approach aims to overcome the limitations of direct radioimmunotherapy, such as long blood half-lives and off-target marrow toxicity.
